Cops managed to arrest the teen suspect, who has admitted to carrying out the appalling attack, according to the Acre government.
Two female staff members were pronounced dead at the scene, while another staff member and a student were rushed to hospital with injuries.
The deadly shooting took place at the public school Instituto Sao Jose in Rio Branco.
Eduardo Rodrigues Cavalcante, who works as a receptionist at a hotel next to the school, said he saw students trying to jump over a wall separating the two buildings.

He recalled: “The wall is six meters (20ft) high, and only one person managed to jump over and take refuge here in the hotel.
“The other people were left on the school roof trying to escape.”
The gun used in the shooting reportedly belonged to the boy’s stepfather, and can be seen lying on the floor in another shocking video of the incident.
The stepdad has also been arrested.
Governor Mailza Assis said: “The state expresses deep solidarity with the victims’ families, the school community of Instituto Sao Jose and all education professionals affected by this incident.”
Last year, two teenagers were killed in a shooting at a school in Ceara state in northeastern Brazil.
Another shooting in Ceara in 2022 left one dead, after a teenager gunned down three classmates.
The most recent attack comes less than one month after a 14-year-old carried out the deadliest school shooting in Turkish history, killing 10 and injuring 19 others.
Isa Aras Mersinli stormed the Ayser Çalık Secondary School in Onikisubat and opened fire in two classrooms before being restrained by two teachers, a canteen worker, and a father of two students, Necmettin Bekci.
Bekci slashed the gunman’s leg with a knife in order to incapacitate him, inadvertently severing the femoral artery and killing him.
It is understood that Mersinli concealed his father’s guns in a backpack to carry out the horrific “indiscriminate” attack on two different classrooms.
Terrifying footage showed screaming children fleeing the building and onto the opposite side of the street.

It came just 24 hours after a former student armed with a pump-action shotgun began firing randomly at a different school in Turkey.
At least 16 people were injured in that attack at the high school in the country’s south east.
Distressing CCTV footage shows the shooter charge towards the school doors as some of his panicked classmates recognise the danger and begin to flee.
The teen then enters the building and looks down at his rifle before beginning his rampage.
Terrified students can be seen running for their lives and barricading themselves in classrooms as the gunman makes his way down the hall.
The attack has left 10 students, four teachers, a canteen employee and a police officer wounded, authorities said.
Governor Hasan Sildak told local media that the 19-year-old shooter took his own life when cops cornered him.
