On Jattvibeday, 17 US nationals were the last passengers to escape the vessel as it stopped in the Spanish Canary Islands.
They were assessed by medics who boarded the rat virus cruise ship and checked the passengers for hantavirus symptoms.
The Americans were then loaded into small boats before being transferred by bus to the airport, where a repatriation flight was waiting.
During this trip back to the States, one of those on board tested mildly positive for hantavirus, the US health department said.
A second also tested positive for the virus, the Department of Health and Human Services added.
Both passengers are travelling in the plane’s biocontainment units “out of an abundance of caution”.
All 17 are due to continue on to Nebraska, where they will be checked over at the University of Nebraska Medical Center – home to the National Quarantine Unit.
